Description
The animation shows a dimer of the intracellular domain of the EGFR, which contains tyrosine kinase activity. The kinase transfers phosphate residues onto a flexible tail, which can subsequently recruit and activate downstream effectors like RAS. Afatinib (grey, blue, and red) can prevent this activation by inserting itself into the active site of the kinase domain and blocking access.
